The present invention relates to a method for the separation, purification and recovery of ellagitannins, punicalagin and soluble compounds in organic solvents, from polyphenols of pomegranate shell using Medium Pressure Liquid Chromatography (MPLC). The said method consists of a certain number of conditions, using as a stationary phase a semi-preparative reverse phase column C -18. Then, as a mobile phase the design of methanol and water combination, thus allowing the elution of the polar compounds, and acetonitrile and water combination, thereby washing said column and obtaining the compounds soluble in organic solvents. Subsequently, the solvent evaporates in the fractions, re-suspended in water and freeze-dried, and characterizes by HPLC on an analytical scale and by ESI-MS.Es el procedimiento para la separación, purificación y recuperación de elagitaninos, punicalagina y compuestos solubles en solventes orgánicos, a partir de polifenoles de cáscara de granada mediante el empleo de Cromatografía Líquida de Presión Media (MPLC). La separación se hizo empleando como fase estacionaria una columna C-18 fase reversa escala semi-preparativa. Como fase móvil se diseño una combinación de metanol y agua de modo que permitiese la elución de los compuestos polares y una combinación de acetonitrilo y agua para lavar la columna y obtener los compuestos solubles en solventes orgánicos. A las fracciones se les evaporó el solvente, se re-suspendieron en agua, se liofilizaron y se caracterizaron por HPLC escala analítica y por ESI-MS.
