The present application describes a method for sequencing oligosaccharides, which enables the primary sequence of an oligosaccharide with unknown structure to be identified, including the composition of same in monosaccharides, the position (regiochemistry) and configuration (stereochemistry) of the glycosidic bonds, the nature and position of the functional modifications, and the branched structure thereof, including in particular identifying the reductive end. Said method of sequencing oligosaccharides comprises the steps of i. fragmentation of the oligosaccharides into disaccharides and monosaccharides while preserving the molecular structure of the constituents as present in the oligosaccharide to be sequenced, ii. separating each previously obtained disaccharide and monosaccharide by mass spectrometry, iii. analysing, by vibrational IR (infrared) spectroscopy, each previously separated disaccharide and monosaccharide, iv. identifying the structure of each disaccharide and monosaccharide by comparing the IR spectra obtained with a set of IR spectra of reference disaccharides and monosaccharides, and v. defining the sequence of the oligosaccharide by combining the structures identified for each disaccharide and monosaccharide.La présente demande décrit un procédé de séquençage d'oligosaccharides, qui permet d'identifier la séquence primaire d'un oligosaccharide de structure inconnue, incluant sa composition en monosaccharides, la position (régiochimie) et la configuration (stéréochimie) des liaisons glycosidiques, la nature et la position des modifications fonctionnelles, et sa structure branchée, dont en particulier l'identification de l'extrémité réductrice.
