The present invention relates to mono-specific and multi-specific polypeptides that specifically bind or interact with CD3. These polypeptides can be, but are not limited to, antibodies, fragments thereof, scFvs, Fabs, di-scFvs single domain antibodies, diabodies, dual variable domain binding proteins and polypeptides containing an antibody or antibody fragments. In one embodiment, a multi-specific polypeptide binds both a T-cell receptor complex on T-cells and a tumor antigen to induce target-dependent T-cell cytotoxicity, activation and proliferation. In a further embodiment, the present invention relates to a CD3 binding polypeptide comprising a CD3 binding domain, wherein the CD3 binding domain has a reduced isoelectric point by at least 0.25 units as compared to the isoelectric point of the binding domain with the amino acid sequence of SEQ ID NO:41 wherein the CD3 binding domain comprises a humanized VH region and a humanized VL region, wherein the humanized VH region and the humanized VL region comprise framework regions and wherein the VH region comprises SEQ ID NO:28, and the VL region comprises SEQ ID NO:34 or an amino acid sequence from 90% to less than 100% identical to SEQ ID NO:34 and wherein the VL region comprises a CDR1 having a sequence of SEQ ID NO: 52, a CDR2 having a sequence of SEQ ID NO: 53, and a CDR3 having a sequence of SEQ ID NO: 54.
