Disclosed are compounds of formula I, R and S enantiomers thereof, and methods for their preparation. A specific example of formula I is (R)-5-(2-(benzylamino)ethyl)-1-(6,8-difluorochroman-3-yl)-1H-imidazole-2(3H)-thione. Also disclosed is a process for the preparation of the individual R and S enantiomers of formula I from the reaction of the terminal primary amine precursor with an aldehyde under reductive alkylation conditions. Compounds of formula I are useful for reducing the hydroxylation of dopamine to noradrenaline, treating anxiety disorders, migraine, cardiovascular disorders, hypertension, chronic or congestive heart failure, angina, arrhythmias, circulatory disorders, Raynauds phenomenon and inhibiting dopamine-beta-hydroxylase.
