Embodiments of the invention stimulate three levels of beta cell physiology:(i) glucosemetabolism, (ii) membrane receptor function, and (iii) transcriptional factorsthat result in the invivo formation of beta cells in the pancreas for the purpose of treatingdiabetes. In certainaspects, the methods include the integration of three levels of cellularphysiology: metabolism,membrane receptor function, and gene transcription. The integration ofmultiple levels of cellularphysiology produces a synergistic effect on beta cell formation.
