Single chain insulin analogs are provided having high potency and specificity for the insulin receptor. As disclosed herein optimally sized linking moieties can be used to link human insulin A and B chains, or analogs or derivatives thereof, wherein the carboxy terminus of the B25 amino acid of the B chain is linked to the amino terminus of the Al amino acid of the A chain via the intervening linking moiety. In on embodiment the linking moiety comprises a polyethylene glycol of 6-16 monomer units and in an alternative embodiment the linking moiety comprises a non-native amino acid sequence derived form the IGF-1 C-peptide and comprising at least 8 amino acids and no more than 12 amino acid in length. Also disclosed are prodrug and conjugate derivatives of the single chain insulin analogs.La présente invention concerne des analogues dinsuline à une seule chaîne très actifs et spécifiques du récepteur à linsuline. Comme décrit ici, des groupes fonctionnels de liaison dune taille optimale peuvent être utilisés pour relier les chaînes A et B de linsuline humaine, ou des analogues ou dérivés de celles-ci, lextrémité carboxy-terminale de lacide aminé B25 de la chaîne B étant reliée à lextrémité amino-terminale de lacide aminé A1 de la chaîne A par lintermédiaire du groupe fonctionnel de liaison interposé. Dans un mode de réalisation, le groupe fonctionnel de liaison comprend un polyéthylèneglycol à 6 à 16 motifs monomères et dans un autre mode de réalisation, le groupe fonctionnel de liaison comprend une séquence dacides aminés non native dérivant du peptide C de lIGF-1, comprenant entre 8 et 12 acides aminés dans sa chaîne. Linvention concerne également des dérivés de type promédicament et conjugué desdits analogues de linsuline à une seule chaîne.
