Methods and systems for synthesizing heparin compounds are provided. A method of chemoenzymatic synthesis of structurally homogeneous low molecular weight heparin with reversible anticoagulant activity is provided. Also disclosed are heparin compounds that have anticoagulant activity, including binding affinity for antithrombin and anti-Xa activity, but no detectable anti-IIa activity. Further provided is a synthesized low molecular weight heparin compound having reversible anticoagulant activity, wherein the anticoagulant activity is reversible by protamine.
