The structural design, preparative methods and chemicalcomposition of two structural formulations of bivalentvaccines against morphine-heroin addiction(morphine-6--hemisuccinyl-EDC-TFCS-tetanus toxoid and3-O--carboxymethylmorphine-EDC-TFCS-tetanus toxoid), aredisclosed. These vaccines are suitable for human use inwhich they are capable of triggering the synthesis ofpolyclonal antibodies against morphine opiate and itsstructural analogue, heroin, through the repeated in vivoadministration of these formulations, in active vaccinationprotocols, in pre-clinical studies in rodents. The activevaccination paradigm through which these immunogens triggera humoral immune response consolidated with a long-termimmunological memory, characterized by the presence of hightiters of specific antibodies against these two drugs ofabuse, is also disclosed. Furthermore, the present inventionreveals the efficacy of these conjugate formulations fortriggering a sustained immunoprotection against morphine andheroin addiction using an intravenous self-administrationparadigm of these two opiate substances in the rodent.Finally, a discussion is also made on the potential futureuse of these immunoconjugates in active vaccinationprotocols for treating both morphine and heroin addiction inthe humans.
