Provided in this application is a novel E. coli O polysaccharide O25B. Further provided in this application are prokaryotic host cells containing enzymes (eg, glycosyltransferases) used in the production of O25B. The host cells provided herein produce an O25B bioconjugate, which bioconjugate comprises O25B conjugated to a carrier protein. Further provided herein are compositions (eg, pharmaceutical compositions) comprising O25B and / or a bioconjugate comprising O25B.
