Disclosed is a nucleic acid molecule comprising a chimeric dengue construct comprising nucleic acid sequences encoding nonstructural proteins and one or more structural proteins from a live, attenuated dengue-2 virus and encoding one or more structural proteins from a dengue-4 virus, wherein the chimeric construct comprises one or more mutations comprising: a mutation in an envelope protein at position 417 of dengue-4, wherein the mutation changes glutamic acid to a positively charged residue a mutation in the capsid protein at position 107 of the dengue-2 virus in the dengue-4/dengue-2 chimera, wherein the mutation changes to an aromatic residue a mutation in NS4A at position 17 in the live, attenuated dengue-2 virus, wherein the live, attenuated dengue-2 virus is a modified dengue-2 PSK-53 virus
