A single recombinant Tetravalent Domain III (rTDIII) protein for use in the detection and or diagnosis of any or all of dengue specific Immunoglobin M anti-dengue IgM and Immunoglobin G anti-dengue IgG antibodies is disclosed. Said protein comprises domain III of envelope protein from all four serotypes of dengue virus, namely, Dengue-virus type-1, Dengue-virus type-2, Dengue-virus type-3 and Dengue-virus type-4, linked with each other through penta glycine linkers and codon optimized for expression in an expression vector. The protein of the present invention shows a very high degree of sensitivity and specificity to anti-dengue IgM and anti-dengue IgG antibodies.
