The present disclosure provides a culture method for improving amount of triterpenoids in inonotus obliquus, and the culture method includes the following steps. First, a strain of the inonotus obliquus is inoculate to a solid medium for producing mycelium of the inonotus obliquus. When an area of the cultivated mycelium of the inonotus obliquus is up to a first predetermined value, a first nutrient solution is added to the solid medium. When the area of the cultivated mycelium of the inonotus obliquus is up to a second predetermined value, a brightness and a temperature of the environment, where the mycelium of the inonotus obliquus is cultured, are lowered until a second nutrient solution is added to the solid medium. When the area of the cultivated mycelium of the inonotus obliquus is up to a third predetermined value, a harvesting step is performed.
