PROCESS FOR PRODUCTION AND EXTRACTION OF WATER SOLUBLE MELANIN BY ACTINOBACTERIUM ABSTRACT: The invention relates to the process for production and extraction of water soluble melanin by an actinobacterium. It particularly relates to the simple process for high yield production of water soluble melanin by an actinobacterium under a standardized submerged bioprocess technology. It particularly relates to the process for high yield production of water soluble melanin by a novel strain of actinobacterium - Streptomyces lusitanus DMZ3 (MTCC 5935) employing submerged bioprocess technology. It particularly focuses on the enhanced production of water soluble melanin in a specific fermentation medium with optimized physicochemical and nutritional variables. It further focuses a unique process for extraction of water soluble melanin from the fermented medium covering several specific physicochemical reactions at various stages. Process for the production and extraction of water soluble melanin comprising: (a) employing the submerged bioprocess technology for culturing of actinobacterium in a specific fermentation medium for production of water soluble melanin and (b) separation of water soluble melanin from the fermented broth. The fermentation medium has effective amount of 0.7% (w/v) tyrosine, 0.7% (w/v), gelatin, 0.35% (w/v) beef extract along with 0.03% (w/v) CuSO4 which are regulated as critical nutritional variables for the production of water soluble melanin. The amount of at least 6.69 g/L of water soluble melanin is produced by Streptomyces lusitanus DMZ3 after 96 h of fermentation. The water soluble melanin produced is separated from fermentation broth by innovative approach.
