ÉPITOPES UNIVERSELS ARTIFICIELS DE CELLULES T AUXILIAIRES QUI FACILITENT LA PRODUCTION CIBLÉE D'ANTICORPS TOUT EN LIMITANT LA RÉPONSE DES CELLULES T DE TYPE INFLAMMATOIRE
The present invention is directed to novel heterologous promiscuous and artificial T helper cell epitopes (Th epitopes) designed to provide optimum immunogenicity of a target antigenic site. The disclosed Th epitopes, when covalently linked to a B cell epitope in a peptide immunogen construct, elicit a strong antibody response to the B cell epitope of the target antigenic site. The Th epitopes are immunosilent on their own, i.e., little, if any, of the antibodies generated by the peptide immunogen constructs will be directed towards the Th epitope, thus allowing a very focused immune response directed to the targeted antigenic site. The heterologous promiscuous Th epitopes provide effective and safe peptide immunogens that do not generate inflammatory, anti-self, cell-mediated immune responses following administration.La présente invention concerne de nouveaux épitopes hétérologues universels et artificiels de cellules T auxiliaires (épitopes Th) conçus pour obtenir une immunogénicité optimale d'un site antigénique cible. Les épitopes Th de l'invention, lorsqu'ils sont liés de manière covalente à un épitope de cellule B au sein d'une construction immunogène peptidique, induisent une forte réponse anticorps contre l'épitope de cellule B du site antigénique cible. Les épitopes Th sont immunosilencieux en eux-mêmes, c'est-à-dire que peu, voire aucun, des anticorps générés par les constructions peptidiques immunogènes ne sera dirigé contre l'épitope Th, permettant ainsi une réponse immunitaire très focalisée dirigée contre le site antigénique ciblé. Les épitopes Th hétérologues universels fournissent des immunogènes peptidiques efficaces et sûrs qui ne génèrent pas de réponses immunitaires à médiation cellulaire de type inflammatoire et auto-immun après administration.
