The present invention relates to a new generation of chimeric antigen receptor (CAR) (referred to as multi-chain CAR) specific for antigen CS1. Such CARs are intended to redirect the specificity and reactivity of immune cells to malignant cells that express the tumor antigen CS1. The alpha, beta and gamma polypeptides that make up these CARs are designed to assemble at near-membrane locations that form a flexible architecture closer to the natural receptor that confers optimal signal transduction. The invention encompasses polynucleotides, vectors, and isolated cells that express them on the surface, encoding the multi-stranded CAR, particularly for use in immunotherapy.
