The present invention relates to a method of preparing a hand cleaning agent composition. The method of the present invention comprises: an extraction step of mixing a natural material with deionized distilled water or a first solvent to obtain a mixed solution and heating the mixed solution to obtain an extract； a filtration step of filtering the extract obtained in the extraction step to obtain a filtrate； a concentration step of concentrating the filtrate filtered in the filtration step； a powder preparation step of freeze-drying the concentrate completed in the concentration step to obtain a powder； and a powder mixing step of mixing the powder prepared in the powder preparation step with a second organic solvent. According to the present invention, the method is able to prepare the natural material in the extraction step by using at least one selected from the group consisting of Phellinus linteus and Houttuynia cordata. Therefore, the method provides a technology of producing a hand cleaning agent which prevents various side effects, possibility of causing a disease on the skin, and the like during using of the hand cleaning agent, which has excellent antibacterial activity, and which can prevent the generation of skin diseases of a user. Further, the method provides a technology of producing a hand cleaning agent having antibacterial performance against resistant bacteria as well as various pathogenic bacteria.
