Herein is reported a method for the generation of multispecific antibodies by a half-antibody exchange reaction between two 2/3-IgGs destabilized in one half by asymmetric perturbing mutations fostering the generation of correctly assemble full length bispecific antibodies. The method can be performed in the absence of reducing agents and does not require hinge region disulfide bonds in the starting 2/3-IgGs.本文中報導一種藉由半抗體交換反應來產生多特異性抗體的方法,該半抗體交換反應在兩個2/3-IgG之間進行,該兩個2/3-IgG的一半藉由不對稱的擾動突變而失去穩定,從而促進正確組裝的全長雙特異性抗體產生。該方法可在還原劑不存在下執行且不需要初始2/3-IgG中存在鉸鏈區二硫鍵。
