TITLE: A PROCESS OF PREPARING PROCESSED HUMAN AMNIOTIC COLLAGEN MEMBRANE FREE FROM CHORION FOR USE AS WOUND DRESS APPLICANT: SREE BALAJI MEDICAL COLLEGE & HOSPITAL, BIHER- BHARATH UNIVERSITY ABSTRACT The present invention shall disclose a process of preparing processed human amniotic collagen membrane free from chorion for use as wound dress for healing wounds, dermal and ocular application. The process comprises of obtaining screened placenta from donors and washed with sterile Hanks™ balanced salt solution under laminar air flow to remove excess blood to make it free from blood clots. Then the chorion membrane from the amniotic membrane is removed under laminar air flow in which amniotic membrane was mechanically detached under laminar air flow from chorion and washed with plurality of times with PBS to allow the malleable stromal layer to swell followed by removal of the swelled stromal layer. Subsequently soaking the membrane into Dulbecco™s Modified Eagle™s medium (DMEM /Eagles Essential Medium (EMEM) supplemented with glutamine, antibiotics and antimycotic solution for predetermined period of time and washed with plurality of times with EMEM/ DMEM containing antibiotics to form chorion free amniotic membrane in which a few left over chorion still attached to the membrane and blood clots were peeled off from the epithelial layer. Further decellularizing the membrane by treating with EDTA or Triton X- 100, proteolytic enzymes) or with 0.5 M NaOH to solubilize and remove unwanted immunogenic molecules and cells from the amniotic membrane to form decellularized amniotic collagen membrane followed by trimming into pieces conforming to area of specification. Finally cross-linking the trimmed amniotic membrane into desired sizes followed by stabilization by physical or chemical method to form the processed dehydrated amniotic collagen membrane.
