The present invention relates to methods and pharmaceutical compositions for the treatment of non-alcoholic fatty liver disease. The inventors showed that autophagy is defective in liver sinusoidal endothelial cells (LSECs) from patients with non-alcoholic steatohepatitis (NASH) and contributes to the main features of this disease. In particular, the present invention relates to a method of treating a non-alcoholic fatty liver disease in a subject in need thereof comprising administering to the subject a therapeutically effective amount of an agent capable of restoring autophagy in the subject's liver sinusoidal endothelial cells.La présente invention concerne des procédés et des compositions pharmaceutiques destinés au traitement de la stéatohépatite non alcoolique. Les inventeurs ont montré que l'autophagie est défectueuse dans les cellules endothéliales sinusoïdales du foie (LSEC) provenant de patients présentant une stéatohépatite non alcoolique (NASH) et contribue aux caractéristiques principales de cette maladie. En particulier, la présente invention concerne un procédé de traitement d'une stéatohépatite non alcoolique chez un sujet en ayant besoin comprenant l'administration au sujet d'une quantité thérapeutiquement efficace d'un agent apte à restaurer l'autophagie dans les cellules endothéliales sinusoïdales du foie du sujet.
