PROBLEM TO BE SOLVED: To provide a method for preparing a conjugate of a cell binding agent such as an antibody stably linked to an effector group or a reporter group to provide a conjugate of high purity and homogeneity in high yields without inter-chain cross-linking and inactivated linker residues.SOLUTION: Provided is a method for preparing a non-reducible, thiol-linked conjugate of the formula C-L-CBA, where C represents an effector molecule (e.g., a cytotoxic agent), L is a linker and CBA is a cell binding agent (e.g., an antibody or a fragment thereof), by utilizing a direct reaction of the thiol-containing cytotoxic agent (e.g., maytansinoids) with a hetero- or homo-bifunctional reagent, (e.g., a cleavable or non-cleavable linker) followed by mixing of the unpurified reaction mixture with a cell binding agent (e.g., an antibody or a fragment thereof), thereby generating the non-reducible, thiol-linked conjugate by a process that is more efficient with a high yield, and is amenable for scale up.【課題】エフェクター基又はリポーター基との抗体のような細胞結合剤の、安定して連結される抱合体の調製方法で、鎖間架橋した及び不活化されたリンカー残基を含まずに高い純度と均質性の高い収率の抱合体を提供。【解決手段】チオール含有の細胞傷害剤（例えば、マイタンシノイド）のへテロ又はホモの２官能性試薬（例えば、切断可能又は切断不能なリンカー）との直接反応を利用し、次いで未精製の反応混合物を細胞結合剤（例えば、抗体又はその断片）と混合し、それによって、さらに効率的であり、高い収率を有し、規模拡大に適する製造過程により式Ｃ－Ｌ－ＣＢＡで表される非還元性のチオール結合の抱合体を生成する方法。（Ｃはエフェクター又はリポーターの分子；Ｌはリンカー；ＣＢＡは細胞結合剤）【選択図】なし
