The present disclosure provides conjugate compounds comprising antibodies and fragments thereof, and more particularly those having therapeutic or diagnostic applications, that are engineered by one or more reactive cysteine residues. Conjugated compounds include, for example, cysteine engineered antibodies or fragments thereof conjugated to chemotherapeutic agents, toxins, and detection labels such as radionuclides or fluorophores. The disclosure also provides methods of using the disclosed conjugate compounds to diagnose or treat mammalian cells, or related pathological conditions, in vitro, in situ, in vitro, and in vivo.
