The present invention relates to primer sets for RT-PCR and nested PCR to specifically detect wheat streak mosaic virus, a composition and a kit comprising the same, and a method for detecting wheat streak mosaic virus using the composition and kit. The detection method according to the present invention can detect wheat streak mosaic virus with detection limit 1,000 times more sensitive than the detection method using the existing antiserum for plant seeds and tissue and respond to all known lineages of wheat streak mosaic viruses, and confirm a positive or negative reaction, thereby being able to be effectively used for industrialization of a RT-PCR detection kit for plant viruses and test for wheat streak mosaic virus in the field of quarantine for imported plants.
