A method of making a cell sheet comprising secondary spheroids, including (a) obtaining cardiosphere-derived cells； (b) cultivating the cardiosphere-derived cells for a first period of time in a first media comprising at least one of an ascorbic acid and an analog thereof, to form secondary spheroids； (c) transferring an amount of the spheroids formed in step (b) into a mold； (d) culturing the secondary spheroids for a second period of time in a second media comprising at least one of the ascorbic acid and an analog thereof, wherein the at least one of the ascorbic acid and an analog thereof is present in an amount effective to promote a formation of an extracellular matrix； and (e) culturing a product obtained in step (d) for a third period of time, in the absence of the at least one of the ascorbic acid and an analog thereof.L'invention concerne une méthode de fabrication d'une feuille cellulaire comprenant des sphéroïdes secondaires, comprenant (a) l'obtention de cellules dérivées de la cardiosphère ； (b) la culture des cellules dérivées de la cardiosphère pendant une première période de temps dans un premier milieu comprenant un acide ascorbique et/ou un analogue de celui-ci, pour former des sphéroïdes secondaires ； (c) le transfert d'une quantité des sphéroïdes formés à l'étape (b) dans un moule ； (d) la culture des sphéroïdes secondaires pendant une seconde période de temps dans un second milieu comprenant de l'acide ascorbique et/ou un analogue de celui-ci, l'acide ascorbique et/ou un analogue de celui-ci étant présent en une quantité efficace pour favoriser la formation d'une matrice extracellulaire ； et (e) la culture d'un produit obtenu à l'étape (d) pendant une troisième période de temps, en l'absence de l'acide ascorbique et/ou d'un analogue de celui-ci.
