PROBLEM TO BE SOLVED: To provide a peptidomimetic macrocycle with improved properties.SOLUTION: The invention provides a biologically active peptidomimetic macrocycle with improved properties compared with a corresponding polypeptide thereof. The invention also provides a method of preparing such a macrocycle and a method of using the same, for example, in therapeutic application. For example, the invention provides a method of increasing the in vivo half-life of a helical polypeptide by installing one or more cross-links.
