The present invention relates to a method for producing polymer microspheres with an increased drug encapsulation efficiency. More particularly, the method includes the steps of producing an inner water phase (W1) including a drug, a thickener, and an aqueous solvent and producing an oil phase including a polymer and a fat-soluble solvent； producing a W1/O emulsion by mixing the inner water phase and the oil phase and increasing viscosity by cooling the W1/O emulsion； producing a W1/O/W2 emulsion by including the W1/O emulsion with increased viscosity, dispersing the W1/O emulsion in an outer water phase (W2) with the temperature maintained to be 18°；C or lower, and mixing the W1/O emulsion and the W2/O emulsion at 180°；C for 10 minutes or more； and drying the W1/O/W2 emulsion produced in the earlier step and recollecting the W1/O/W2 emulsion. The method of the present invention can be effective for producing a slow release drug as a drug encapsulation efficiency is increased by a process of maintaining a certain temperature for a certain time to solidify the oil phase, and the polymer microspheres with a high drucg content can be produced.본 발명은 약물 봉입효율을 증가시킨 고분자미립구 제조방법에 관한 것으로, 보다 상세하게는 약물, 증점제 및 수성 용매가 포함된 내부수상(W1)을 제조하고, 중합체 및 지용성 용매가 포함된 오일상(O)을 제조하는 단계； 상기 단계에서 제조된 내부수상과 오일상을 혼합하여 W1/O에멀젼을 제조하고, 상기 제조된 W1/O에멀젼을 냉각하여 점도를 증가시키는 단계； 상기 단계에서 점도가 증가된 W1/O 에멀젼을 수용성 용액이 포함되며, 온도가 18℃ 이하로 유지된 외부수상(W2)에 분산시키고 온도를 18℃이하로 10분 이상 유지하면서 혼합하여 W1/O/W2 에멀젼을 제조하는 단계； 및 상기 단계에서 제조된 W1/O/W2 에멀젼을 건조하고 회수하는 단계를 포함하는 고분자미립구 제조방법에 관한 것이다. 본 발명의 방법은 오일상을 고형화 시키는 과정을 특정 온도를 특정 시간 이상 유지하는 과정을 통하여 약물 봉입효율을 증가시키고, 높은 약물함량을 가지는 고분자미립구를 제조할 수 있어, 서방출형 약물 제조에 효과적이다.
