The present invention relates to a single domain antibody capable of specifically binding to the PAS-B domains of transcription factors HIF-1α and HIF-2α activated in hypoxia, and to antigen-binding fragments thereof. The single domain antibody specific for the PAS-B domain of HIF-1α and HIF-2α according to the present invention binds to the domain where HIFs α and HIF β (ARNT) interact to inhibit HIFs/ARNT complex formation, as well as HIF1α. The /ARNT and HIF2α/ARNT complex can be usefully used to suppress the expression of genes that are specifically transcribed, and can be used as a therapeutic agent for diseases such as cancer.본 발명은 저산소 상태에서 활성화되는 전사인자(transcription factor) HIF-1α와 HIF-2α의 PAS-B 도메인에 특이적으로 결합할 수 있는 단일 도메인 항체 및 이의 항원 결합 단편에 관한 것이다.본 발명에 따른 HIF-1α와 HIF-2α의 PAS-B 도메인에 특이적인 단일 도메인 항체는 HIFs α와 HIF β(ARNT)가 상호작용하는 도메인에 결합하여 HIFs/ARNT 복합체 구성을 저해할 뿐만 아니라 HIF1α/ARNT 및 HIF2α/ARNT 복합체가 특이적으로 전사되는 유전자의 발현을 억제시키는데 유용하게 사용할 수 있으며, 이를 이용하여 암과 같은 질병 치료제 등으로 활용할 수 있다.
