A multi-layered liposome structure is a multi-layered spheroid structure including a core layer, a first outer layer, and a second outer layer respectively made of poly(r-glutamic acid)(r-PGA), phospholipids, and poly(r-glutamic acid) hydrogel. A preparation method of the multi-layered liposome structure includes the following steps: a phospholipid film preparation step, a mixing step, and a wrapping step. The phospholipid film preparation step uses a rotary vacuum condensation process to remove solvent contained in the phospholipid solution so that the phospholipids forms a phospholipid thin film on a wall of container. The mixing step adds r-PGA solution into the container having said phospholipid thin film to obtain liposome solution. The wrapping step adds r-PGA hydrogel solution into the liposome solution that is subjected to homogenization to have a hydrogel layer wrapped around an outer layer of the liposome.一種多層微脂粒結構,為一多層球體結構,係包含一中心層、一第一外層及一第二外層,分別為聚麩胺酸、磷脂質及聚麩胺酸水膠。一種多層微脂粒結構之製備方法,其步驟包含:一磷脂質薄膜製備步驟、一混合步驟及一包覆步驟。該磷脂質薄膜製備步驟,以一減壓迴旋濃縮方式去除一磷脂質溶液中的溶劑,使該磷脂質於該容器的瓶壁上形成一層磷脂質薄膜;該混合步驟,將一聚麩胺酸(γ-PGA)溶液加入含有該磷脂質薄膜的容器中,以獲得一微脂粒溶液;及該包覆步驟,將一聚麩胺酸水膠溶液加入該微脂粒溶液中並進行均質,使該微脂粒外層進一步包覆有一水膠層。
