PROBLEM TO BE SOLVED: To provide a method for producing a composite type cultured skin which is excellent in flexibility, hardly forms a scar tissue, when transplanted, and has an elastic fibrous tissue layer between an epidermic layer and a dermal layer, in a short period.
SOLUTION: The method for producing the cultured skin having an elastic fibrous tissue layer between an epidermal cell layer and a dermal cell layer, comprises a process 1 for sowing and culturing fibroblasts on a first porous substrate comprising collagen and having an average pore size of 50 to 150 &mum in density of &ge1×10
4 fibroblasts/cm2 , a process 2 for superposing a second porous substrate comprising collagen and having an average pore diameter of 1 to 30 &mum on the first porous substrate having the fibroblasts sowed thereon and sowing and culturing fibroblasts on the second porous substrate in density of &ge1×103 fibroblasts/cm2 , and a process 3 for superposing a third porous substrate comprising collagen and having an average pore diameter of 1 to 30 &mum on the second porous substrate having the fibroblasts sowed thereon and sowing kelatinocytes on the third porous substrate in density of &ge1×104 fibroblasts/cm2 .COPYRIGHT: (C)2010,JPO&INPIT
