PROBLEM TO BE SOLVED: To provide an amphetamine dosage form having therapeutically effective abuse resistant properties, or the amphetamine dosage form having sustained release and continuous effect of treatment.SOLUTION: The invention relates to compounds, compositions and methods of using the same comprising a chemical moiety covalently attached to amphetamine. The compounds and compositions are useful for reducing or preventing abuse and overdose of amphetamine. The compounds and compositions find particular use in providing an abuse-resistant alternative treatment for certain disorders, such as attention deficit hyperactivity disorder (ADHD), ADD, narcolepsy, and obesity. Oral bioavailability of amphetamine is maintained at therapeutically useful doses. At higher doses bioavailability is substantially reduced, thereby providing a method of reducing oral abuse liability. Further, compounds and compositions decrease the bioavailability of amphetamine by parenteral routes, such as intravenous or intranasal administration, further limiting their abuse liability.【課題】治療的に有効な乱用抵抗性のアンフェタミン剤形あるいは徐放性および持続的な治療効果を提供するアンフェタミン剤形を提供すること。【解決手段】本発明は、アンフェタミンに共有結合している化学的部分を含む化合物、組成物およびそれらの使用法を記載する。これらの化合物および組成物は、アンフェタミンの乱用および過量の減少または防止に有用である。これらの化合物および組成物は、注意力欠陥多動性障害（ＡＤＨＤ）、ＡＤＤ、睡眠発作および肥満などの一定の障害に対する乱用抵抗性の代替治療の提供において特に使用される。アンフェタミンの経口生物学的利用能は、治療的に有用な用量において維持される。高用量では生物学的利用能は大きく減少し、そのため経口乱用傾向を低下させる方法が提供される。さらに、本発明の化合物および組成物は、静脈内投与または鼻腔内投与などの非経口経路によるアンフェタミンの生物学的利用能を低下させて、さらにそれらの乱用傾向を制限する。【選択図】 なし
