PURPOSE: Eugenol is provided to be directly applied onto a lesion and be used as a pharmaceutical composition which selectively treats mechanical allodynia.CONSTITUTION: A pharmaceutical composition for treating mechanical allodynia contains eugenol or a pharmaceutically acceptable salt thereof in a concentration that is lower than the concentration that suppresses VGSCs(Voltage-Gated Na Channels) of a nerve cell. The concentration of eugenol or a pharmaceutically acceptable salt thereof is less than 2 mM. The composition additionally contains a pharmaceutically acceptable carrier. The composition selectively suppresses an HCN channel of the nerve cell. A transdermal formulation for treating mechanical allodynia contains the composition.COPYRIGHT KIPO 2013본 발명은 유제놀 또는 이의 약학적으로 허용가능한 염이 VGSCs(voltage-gated Na channels)를 억제하는 농도보다 낮은 농도로 제공되는 기계적 이질통의 선택적 치료용 약학적 조성물, 과분극-활성 전류(hyperpolarization-activated current Ih) 차단용 약학적 조성물, 및 상기 조성물을 포함하는 기계적 이질통 치료용 경피투여형 제제에 관한 것이다.본 발명의 유제놀은 VGSCs(voltage-gated Na channels)를 억제하는 농도보다 낮은 농도로 투여되어 cAMP나 G-단백질 연결 수용체와는 독립적인 기전으로 과분극-활성 전류(hyperpolarization-activated current Ih)를 억제하여 기계적 이질통을 선택적으로 완화시킬 수 있다. 따라서, 본 발명의 유제놀은 VGSCs를 억제하는 농도보다 낮은 농도로 제공되도록 경피투여형 제제로 제형화되어 환부에 직접 적용하여 기계적 이질통을 선택적으로 치료할 수 있는 약학적 조성물로 유용하게 사용될 수 있다.
