The present invention relates to high efficiency encapsulation of hydrophilic materials in the hydrophilic space of monolayer liposomes. High efficiency encapsulation is achieved by the use of polyhydric alcohols selected from propylene glycol or glycerin to dissolve the hydrophobic compounds forming the lipid bilayer of liposomes. The present invention provides monolayer liposomes (UL), and methods for their preparation by low temperature extrusion.
