FIELD: biochemistry.SUBSTANCE: disclosed is a method obtaining a purified cell-binding agent - cytotoxic agent conjugate (embodiments). In the first embodiment, the method comprises bringing the cell-binding agent into contact with a cytotoxic agent to form a first mixture, bringing the first mixture into contact with a linker-containing bifunctional crosslinking agent to form a second mixture, bringing the second mixture into contact with a polyvinyl difluoride membrane and treating the second mixture to obtain a purified mixture of the cell-binding agent-cytotoxic agent conjugate. In a second embodiment, the method comprises bringing the cell-binding agent into contact with a linker-containing bifunctional cross-linking agent to produce a first mixture, treating the first mixture to produce a purified first mixture, conjugating a cytotoxic agent with bound linker-containing cell-binding agents to obtain a second mixture, bringing the second mixture into contact with a polyvinyl difluoride membrane and treating the second mixture to obtain a purified mixture of the cell-binding agent-cytotoxic agent conjugate.
