SOLUTION: A mutation is generated in the nucleic acid sequence of a parent antibody so as to encode a cysteine engineered antibody, and one or more amino acid residues are replaced with cysteine, a cysteine engineered antibody is expressed, and a cysteine engineered antibody is used. A cysteine engineered antibody that comprises a free cysteine amino acid in the heavy or light chain prepared by isolation.
