Vynález se týká sloučenin podobných glukagonu typu 1 fúzovaných na proteiny, které mají za účinek prodloužení poločasu životnosti peptidů in vivo. Heterologní fúzní protein obsahuje první polypeptid s N-zakončením a C-zakončením fúzovaný na druhý polypeptid s N-zakončením a C-zakončením, kde první polypeptid je GLP-1 sloučenina, schopná vázat se na GLP-1 receptor a indukovat signalizaci přes tento receptor, a druhý polypeptid je vybraný z lidského albuminu, analogů lidského albuminu, a fragmentů lidského albuminu, a kde C-zakončení prvního polypeptidu je fúzováno na N-zakončení druhého polypeptidu, přičemž fúzní protein je biologicky aktivní a má delší poločas životnosti v plazmě než samotná GLP-1 sloučenina. Tyto fúzní proteiny mohou být použity pro léčbu diabetu mellitu nezávislého na inzulinu, stejně jako řady dalších chorob.The present invention relates to glucagon-like-1 compounds fused to proteins that have the effect of extending the in vivo half-life of the peptides. A heterologous fusion protein comprises a first polypeptide with a N-terminus and a C-terminus fused to a second polypeptide with a N-terminus and a C-terminus wherein the first polypeptide is a GLP-1 compound that is able to bind and induce signaling through the GLP-1 receptor and the second polypeptide is selected from the group consisting of a) human albumin； b) human albumin analogs； and c) fragments of human albumin, and wherein the C-terminus of the first polypeptide is fused to the N-terminus of the second polypeptide, wherein the fusion protein is biologically active and has a longer plasma half-life than the GLP-1 compound alone. These fusion proteins can be used to treat non-insulin dependent diabetes mellitus as well as a variety of other diseases.
