The present invention provides multispecific binding proteins that bind to NKG2D and CD16 receptors in natural killer cells and tumor-associated antigens selected from 5T4, GPNMB, FR-α, PAPP-A and GPC3. Such proteins can associate with more than one NK activating receptor and block the binding of the natural ligand to NKG2D. In certain embodiments, the protein may stimulate NK cells in humans.
