NANOAGGREGATES SENSITIVE TO PH AND REDUCTION, A PROCESS FOR THE PREPARATION THEREOF, AND A PHARMACEUTICAL COMPOSITION FOR TREATING MALIGNANT TUMOR COMPRISING THE SAME
PURPOSE: A nanoaggregate is provided to deliver a hydrophilic anticancer gene and a hydrophobic chemotherapeutic agent into a cell using one drug delivery system and to deliver an anticancer agent with high efficiency. CONSTITUTION: A nanoaggregate contains 1 micelle unit containing a hydrophobic chemotherapeutic agent conjugated to one end of a biocompatible non-ionic polymer and a hydrophilic anticancer gene conjugated to the other end of the biocompatible non-ionic polymer. The hydrophobic chemotherapeutic agent is conjugated to the biocompatible non-ionic polymer through a reduction-sensitive linker which is isolated under the presence of a reducing agent. The hydrophilic anticancer gene is conjugated to the biocompatible non-ionic polymer through a pH-sensitive linker which is isolated at pH 4-6. The 1 micelle unit has a structure of chemical formula 1. A method for preparing the nanoaggregate comprises: conjugating the hydrophobic chemotherapeutic agent at one end of the biocompatible non-ionic polymer through the reduction-sensitive linker; conjugating the hydrophilic anticancer gene to the other end through the pH-sensitive linker; and dispersing the complex containing the hydrophilic anticancer gene and the hydrophobic chemotherapeutic agent for self-assembling.본 발명은 생체 적합성 비이온성 고분자의 일 말단에 소수성 화학요법제가 결합되고 다른 말단에 친수성 항암 유전자가 결합된 복합체인 1 미셀 단위체를 포함하는 미셀 형태의 나노 응집체로서, 상기 소수성 화학 요법제는 환원제의 존재 하에서 분리되는 환원 민감성 링커를 경유하여 상기 생체 적합성 비이온성 고분자에 결합되고, 상기 친수성 항암 유전자는 pH 4 ~ 6에서 분리되는 pH 민감성 링커를 경유하여 상기 생체 적합성 비이온성 고분자에 결합된, 나노 응집체, 그 나노 응집체의 제조방법, 및 그 나노 응집체를 포함하는 악성 종양 치료용 약학 조성물을 제공한다.
