This invention provides methods and compositions for in vivo incorporation ofunnatural amino acids. Provided are translation systems, cells containingtranslation systemsand compositions including proteins with unnatural amino acids. A translationsystem of thisinvention may comprise an orthogonal tRNA that recognizes at least oneselector codon andan orthogonal amino acyl tRNA synthetase which preferentially aminoacetylatesthe tRNAwith an unnatural amino acid. Such a translation system may be used to producea proteincomprising the unnatural amino acid.
