Nucleases and methods of using these nucleases for altering CFTR genes and producing cell and animal models. Disclosed herein are methods and compositions for altering the CFTR locus. Models for studying the function of CF genes (eg, CFTR), models for CF drug discovery and for treating CF, as well as methods of making and using these model systems are also described.
