The invention provides a method for the preparation of fentanyl comprising: (a) reacting 4-piperidone hydrochloride (NPP) with aniline in presence of reducing environment to produce 4-anilinopiperidine (4- ANPP), (b) alkylating/reacting the 4- ANPP as obtained from step (a) with phenethyl halide under reflux conditions in highly alkaline medium to give 4-anilino-N-phenethylpiperidine, and (c) converting the said 4-anilino-N-phenethylpiperidine to fentanyl by reacting with propionyl chloride in presence of halogenated hydrocarbons then isolating fentanyl by solvent extraction and purified by crystallization from petroleum ether (60-80 °C).L’invention a trait à un procédé de préparation de fentanyle comprenant les étapes consistant à : (a) faire réagir de l’hydrochlorure de 4-pipéridone (NPP) avec de l’aniline en présence d’un milieu réducteur pour produire de la 4-anilinopipéridine (4-ANPP), (b) alkyler/faire réagir la 4-ANPP obtenue à l’étape (a) avec un halogénure de phénéthyle porté au reflux dans un milieu hautement alcalin pour donner de la 4-anilino-N-phénéthylpipéridine, et (c) transformer ladite 4-anilino-N-phénéthylpipéridine en fentanyle en la faisant réagir avec du chlorure de propionyle en présence d’hydrocarbures halogénés, puis en isolant le fentanyle par extraction avec un solvant et en effectuant une purification par cristallisation au moyen d’éther de pétrole (60-80°C).
