PROBLEM TO BE SOLVED: To provide a device and a method for monitoring the Ca
2+ kinetics in many cells inoculated into a scaffold or cells in the whole regenerated tissue rather than single cell unit, for the purpose of the short-term assessment and determination of stimulation conditions promoting tissue regeneration from cells.SOLUTION: The method for monitoring intracellular Ca
2+ kinetics in cells inoculated into a scaffold or in a tissue regenerated from the above cells is provided, comprising the step (i) of bringing a cell membrane-permeable Ca2+ -sensitive fluorescent probe into contact with cells inoculated into a scaffold in a chamber for culturing a regenerated tissue or cells in the regenerated tissue to effect uptake of the probe into the cells, the step (ii) of stimulating the cells inoculated into the scaffold or the regenerated tissue and irradiating the above cells or the regenerated tissue with exciting rays to detect fluorescence emitted from the Ca2+ -sensitive fluorescent probe bound to Ca2+ increased in the cells, and the step (iii) of measuring the change in Ca2+ concentration in the cells inoculated into the scaffold or the cells in the regenerated tissue, based on the fluorescence detected.COPYRIGHT: (C)2009,JPO&INPIT
