The present invention relates to a pharmaceutical composition for preventing and treating neural autoimmune diseases, comprising a mixed extract of Coptidis rhizome, Scutellariae Radix, and Rhei Rhizoma as an active ingredient. In particular, the mixed extract of the present invention, in an animal model of experimental autoimmune encephalomyelitis (EAE), not only alleviates the severity of a neurological condition of the disease and weight loss, but also down-regulates demyelination and activities of neuroglia (microglia and astrocytes) in spleen； down-regulates the mRNA expressions of inflammatory cytokinases (TNF-α, IL-1ß), chemokines (RANTES, MCP-1, MIP-1α) and iNOS； down-regulates the mRNA expressions of endothelial adhesion proteins (ICAM-1, VCAM-1) and junction factors (claudin-3, claudin-5, ZO-1)； and down-regulates NF-kB signalling. These effects were related to a mechanism of down-regulating the activity of Th1 cells by increasing the activity of Treg cells. Accordingly, the mixed extract of the present invention can be utilized as food and drug products that can prevent the emergence of neural autoimmune diseases including multiple sclerosis, or can treat such neural autoimmune diseases.본 발명은 황련(Coptidis rhizome), 황금(Scutellariae Radix) 및 대황(Rhei Rhizoma)의 혼합추출물을 유효성분으로 함유하는 신경계 자가면역질환의 예방 및 치료용 약학적 조성물 및 건강 기능 식품에 관한 것으로, 구체적으로 본 발명의 혼합추출물은 실험적 자가면역성 뇌척수염(EAE) 동물모델에서 질병의 신경학적인 장애의 정도와 체중의 감소를 개선할 뿐만 아니라, 척수에서 탈수초화와 신경아교세포(미세아교세포와 별아교세포)의 활성을 억제하였으며, 염증성 사이토카인(TNF-α, IL-1ß), 케모카인(RANTES, MCP-1, MIP-1α) 및 iNOS의 mRNA의 발현, 내피세포 부착단백질(ICAM-1, VCAM-1)과 연접인자(claudin-3, claudin-5, ZO-1)의 mRNA 발현과 NF-kB 신호기전을 억제하였다. 이러한 효과는 Treg 세포의 활성을 증가시켜서 Th1 세포의 할성을 억제하는 기작과 관련이 있었다. 그러므로 본 발명의 혼합추출물을 다발성 경화증을 포함한 신경계 자가면역질환의 발생을 예방하거나 치료할 수 있는 식의약품으로 활용할 수가 있다.
