Disclosed is a method for preparing a drug-loaded microsphere, the microsphere prepared using the method and the use thereof, wherein the method comprises: mixing a surfactant and water to prepare the aqueous phase mixing the drug and drug carrier with the polymer material, a first solvent and a second solvent to prepare the oil phase and adding the oil phase into the aqueous phase and mixing same, wherein the first solvent and the second solvent are different, and the second solvent and the first solvent are miscible, the second solvent is at least one of a C1-C3 alcohol, a C1-C3 fatty acid, a C1-C3 fatty acid ester, a C1-C2 ether and acetonitrile, and the amount of the first solvent is greater than the amount of the second solvent.Linvention concerne un procédé de préparation dune microsphère chargée en médicament, la microsphère préparée par ce procédé et lutilisation de cette dernière, le procédé consistant : à mélanger un agent tensioactif et de leau pour préparer la phase aqueuse à mélanger le médicament et lexcipient du médicament avec le matériau polymère, un premier solvant et un second solvant pour préparer la phase huileuse et à ajouter la phase huileuse dans la phase aqueuse et mélanger ces dernières, le premier solvant et le second solvant étant différents, et le second solvant et le premier solvant étant miscibles, le second solvant étant au moins lun des éléments du groupe constitué par les alcools en C1-C3, les acide gras en C1-C3, les esters dacide gras en C1-C3, les éthers en C1-C2 et lacétonitrile, et la quantité du premier solvant étant supérieure à la quantité du second solvant.
