PROBLEM TO BE SOLVED: To provide methods and compositions for treating subjects suffering from iduronate 2-sulfatase deficiency in the CNS.SOLUTION: Disclosed herein is a method for treating iduronate 2-sulfatase deficiency in the central nerve system of a subject in need thereof, comprising systemic administration of a therapeutically effective amount of a fusion antibody having an iduronate 2-sulfatase activity. The fusion antibody comprises: (a) a fusion protein comprising an amino acid sequences of an immunoglobulin heavy chain and an amino acid sequence of an iduronate 2-sulfatase, where the amino acid sequence of the iduronic acid-2-sulfatase is covalently linked to the carboxy terminus of the amino acid sequence of the immunoglobulin heavy chain； and (b) an immunoglobulin light chain, where the fusion antibody crosses the blood brain barrier (BBB) and catalyzes the hydrolysis of the 2-sulfate group of L-iduronate 2-sulfate unit of dermatan sulfate, heparan sulfate or heparin.SELECTED DRAWING: None【課題】ＣＮＳにおけるイズロン酸－２－スルファターゼ欠乏症を患っている対象を治療するための方法及び組成物の提供。【解決手段】治療を必要とする対象の中枢神経系におけるイズロン酸－２－スルファターゼ欠乏症を治療する方法であって、イズロン酸－２－スルファターゼ活性を有する融合抗体の治療的有効量の対象への全身投与。該融合抗体は、（ａ）免疫グロブリン重鎖及びイズロン酸－２－スルファターゼのアミノ酸配列を含む融合タンパク質であって、イズロン酸－２－スルファターゼのアミノ酸配列が免疫グロブリン重鎖のアミノ酸配列のカルボキシ末端に共有結合している融合タンパク質；並びに（ｂ）免疫グロブリン軽鎖；を含み、融合抗体が血液脳関門（ＢＢＢ）を通過し、かつ、デルマタン硫酸、ヘパラン硫酸又はヘパリンのＬ－イズロン酸－２－硫酸単位の２－硫酸基の加水分解を触媒する。【選択図】なし
