The present invention relates to a cytosol-penetrating antibody and a use thereof. Specifically, the present invention relates to a light chain variable region and/or a heavy chain variable region which is positioned in a cytosol with drastically improved endosomal escape efficiency by grasping a structural mechanism causing an endosomal escape which is an escape from an endosome to a cytosol after cellular internalization through a cell membrane protein of a living cell； a cytosol penetrating antibody including the same； a method for manufacturing the same； and a use thereof. The cytosol-penetrating antibody or an antigen-binding fragment thereof penetrates into a living cell to be in cytosol without a specific external protein delivery system.본 발명은 세포질 침투 항체 및 이의 용도에 관한 것으로, 구체적으로 살아있는 세포의 세포막 막단백질을 통해 내재화(cellular internalization) 후 엔도좀(endosome)에서 세포질(cytosol)로 탈출하는 엔도좀 탈출을 일으키는 구조적인 기작을 규명하여, 엔도좀 탈출 효율성이 현저히 향상된 세포질에 위치하는 경쇄 가변영역 및/또는 중쇄 가변영역, 이를 포함하는 세포질 침투 항체, 이의 제조방법 및 이의 용도에 관한 것이다.
