The present disclosure relates to a compound referred to herein as mitoketocin, which binds to at least one of ACAT1 / 2 and OXCT1 / 2 and inhibits mitochondrial ATP production. Methods of screening compounds for mitochondrial inhibitory and anti-cancer properties are disclosed. Also described are methods of using mitoketocin to prevent or treat cancer, bacterial infections, and pathogenic yeast, as well as methods of using mitoketocin to provide anti-aging effects. Specific mitoketocin compounds are also disclosed.
