A method for determining an analyte concentration in a physiological sample, the method comprising: (a) introducing the physiological sample into an electrochemical cell to initiate a reaction with a reagent in the cell, the electrochemical cell comprising the first and second electrodes in a separate relationship, the reagent layer comprised an enzyme and a mediator； (b) apply a first electrical potential that has a first polarity to the reaction cell and measure the cell charge as a function of time to obtain a first time-load transient； (c) apply a second electrical potential that has a second polarity to the cell, in which the second polarity is opposite to the first polarity, and measure the cell current as a function of time to obtain a second time transient- load； (d) calculate a first load value during a first time interval of the second transitory load time； (e) calculate a second load value during a second time interval of the second time-load transient and a third load value during a third time interval of the first time-load transient, where the first, second and third time intervals are determined empirically so that the concentration of analyte determined is accurate within ± 10% of a reference measurement, and has a variation of 2% in 1σ； (f) calculate a relationship comprising the second load value divided by the third load value； and (g) multiply the first load value minus a background value by the ratio to derive the analyte concentration.Un metodo para determinar una concentracion de analito en una muestra fisiologica, comprendiendo el metodo: (a) introducir la muestra fisiologica dentro de una celda electroquimica para iniciar una reaccion con un reactivo en la celda, la celda electrquimica comprendiendo el primer y segundo electrodos en una relacion separada, la capa de reactivo comprendinedo un enzima y un mediador； (b) aplicar un primer potencial electrico que tiene una primera polaridad a la celda de reaccion y medir la carga de la celda como
