pharmaceutical formulation of nanoparticles and their preparation methods and uses. compositions, which are storage stable, and a method of producing pharmaceutical-based nanoparticle formulations for clinical use in photodynamic therapy comprising a hydrophobic photosensitizer, poly (lactic-co-glycolic) acid and stabilizing agents are provided. These nanoparticle pharmaceutical formulations provide therapeutically effective amounts of photosensitizer for parenteral administration. In particular, tetrapyrrole derivatives may be used as photosensitizers, the efficacy and safety of which are enhanced by such nanoparticle formulations. The method of preparing plga-based nanoparticles under sterile conditions is also taught. In one preferred embodiment of the present invention, plga-based nanoparticles have an average particle size of less than 500 nm and the photosensitizer is temoporphine, 5,10,15,20-tetrakis (3-hydroxyphenyl) chlorine (mthpc). In another embodiment, the 2,3-dihydroxy-5,10,15,20-tetrakis (3-hydroxyphenyl) chlorine (mthpd-oh) photosensitizer is formulated as a nanoparticle for parenteral administration. however, in another preferred embodiment, the photosensitizer is 5,10,15,20-tetrakis (3-hydroxyphenyl) porphyrin (mthhp).
