PROBLEM TO BE SOLVED: To provide a test method or the like that can assay life and death about Acanthamoeba (cyst) easily and reliably.SOLUTION: A test method includes: (1) a cyst culture preparation step of preparing a cyst culture by growing Acanthamoeba on a solid medium for growth obtained by adding a substance serving as bait of trophozoite ameba, and by making a cyst exist by bringing all Acanthamoeba into cyst configuration, with degradation of the breeding condition due to the decrease of the bait (2) an exposure step of immersing the cyst culture in a test liquid containing a test substance (3) a trophozoite derivation step of deriving the trophozoite ameba from the cyst configuration, by placing the cyst culture passed through the exposure step on a nutrient agar and culturing it and (4) an evaluation step of calculating a de-cyst rate by counting a de-cyst count showing the trophozoite ameba grown from the cyst configuration and being the number of de-cyst traces, and a non-de-cyst count being the number of non-de-cysts remaining as the cyst configuration, after a constant period of time elapses from the implementation of the trophozoite derivation step.COPYRIGHT: (C)2015,JPO&INPIT【課題】アカントアメーバ(・シスト)についての生死を容易且つ確実に定量できる試験方法などを提供する。【解決手段】(1)栄養体アメーバの餌となる物質を添加した増殖用の固形培地上でアカントアメーバを増殖させ、前記餌の減少による飼育条件の悪化に伴い、全てのアカントアメーバをシスト形態とさせることで、シストを存在させたシスト培地を作成するシスト培地作成工程、(2)被験物質を含有した試験液中に、前記シスト培地を浸漬する暴露工程、(3)前記暴露工程を経たシスト培地を栄養寒天培地上に載置して培養することで、シスト形態から栄養体アメーバを誘導する栄養体誘導工程、及び(4)前記栄養体誘導工程の実施から一定時間の経過後、シスト形態から栄養体アメーバとなったことを示す脱シスト跡の個数である脱シスト数と、シスト形態のままの未脱シストの個数である未脱シスト数とを計数し、脱シスト率を算出する評価工程を備えた。【選択図】図1
