The invention features a oligonucleotide of formula I, or pharmaceutically acceptable salts, or prodrugs thereof:which are capable of inducing an anti-viral or an antibacterial response, in particular, the induction of type I IFN, IL-18 and/or IL-1β by binding to RIG-I. The invention relates to methods of making and using modified oligonucleotide comprising at least one triphosphate or analogs thereof.
