The present application discloses cell clusters that resemble the function and characteristics of endogenous islets, and methods for making and using such cell clusters. The present disclosure is an in vitro cell cluster containing at least one unnatural pancreatic β-cell that exhibits a glucose-stimulated insulin secretory response in vitro when exposed to glucose loading, the cell cluster being unsorted. Cell clusters, which are at least about 35% of cells expressing NKX6.1 and C-peptides； at least about 70% of cells expressing chromogranin A； up to about 2% of cells expressing SOX2； Alternatively, a cell cluster containing up to about 10% of cells expressing SOX9 is provided.
